At that point, the game was already well into the production process and Sans and Papyrus’s characters were fully fleshed out.
The Kickstarter video was released in 2013 and his tweet regarding both Sans and Papyrus’s sexualities was made in 2014. Recent examples include the Gaster update and the PS4 release of UnderTale which has added content including new dialogue in the code despite Toby saying he wants to end UnderTale or has stopped working on it previously. Toby is a known troll so for him to say something isn’t canon means something MUST be partially canon. It was CLEARLY a joke post, meaning some of his posts CAN be cited as canon.
